Optimized Single-Step Recovery of Lipophilic and Hydrophilic Compounds from Raspberry, Strawberry and Blackberry Pomaces Using a Simultaneous Ultrasound-Enzyme-Assisted Extraction (UEAE)

An ultrasound-enzyme-assisted extraction (UEAE) was optimized to extract, simultaneously, the hydrophilic and lipophilic compounds from three berry pomaces (raspberry, strawberry blackberry). First, an enzyme screening designated a thermostable alkaline protease as most suitable recover, in aqueous medium, highest yields of polyphenols oil efficient way. Secondly, selected coupled ultrasounds (US) sequential simultaneous combinations. The US–alkaline combination one-single-step process then by definitive design (DSD). parameters were: US amplitude, 20% (raspberry pomace) or 70% (strawberry blackberry pomaces); pH, 8; E/S ratio, 1% (w/w); S/L 6% (w/v); time, 30 min; temperature, 60 °C. Compared conventional extractions using organic solvents, UEAE extracted all polyphenols, with around 75% active (measured DPPH● method) up initial pomaces. Characterized were rich polyunsaturated fatty acids (PUFAs), tocols phytosterols. polyphenolics analyzed UPLC-MS/MS; characteristic ellagitannins Rosaceae family (sanguiin H-6 agrimoniin, sanguiin H-10, …) ellagic acid conjugates found major components.
